Abaloparatide is a synthetic analog of human parathyroid hormone-related protein (PTHrP) consisting of the N-terminal 34 amino acids with specific modifications. It is approved for the treatment of osteoporosis in postmenopausal women at high risk of fracture, and in men with primary or hypogonadal osteoporosis. Compared to teriparatide, abaloparatide demonstrates more transient receptor activation with less sustained cAMP signaling, potentially offering an improved bone formation-to-resorption ratio and a favorable cardiovascular safety profile.
